To "test drive" separations with a 96 well format, Atoll offers a Starter Kit which enables you to develop a screening protocol on a single row of 8 columns, which can be then transferred to a full 96 plate.

This Starter Kit can be seen as the simplest chromatographic system in the world.

The Starter Kit consists of an 8-row of columns containing different media  stacked into a slider rack. This sits on a frame on top of a UV-readable 96-wellplate (e.g. UV-Star Greiner).

How to operate the Kit?
Using a positive displacement pipette set to dispense mode, adjust the speed to about 0.5 ml per minute.

  • 1. Prepare the appropriate buffers.
  • 2. Condition all columns with starting buffer
  • 3. Apply sample(s)
  • 4. Elute with appropriate buffer into adjacent wells by shifting the 8-row along the 12 positions
  • 5. Read results in a plate reader

By this method 12 binding or elution step conditions can be checked.
To optimise buffer/solvent usage and preparation, please use for each chromatographic mode, a corresponding Kit

MediaScout® PipetColumns are of 5mm internal diameter and up to 10mm bed height. They contain each up to 0.2 ml of the chromatography resins, packed to defined density, corrected for each individual resin material, in order to replicate conditions in larger columns. Columns are run stepwise in sequence. Flow is controlled by a positive displacement pipette. Columns are designed for resin screening and for the early stages of method scouting. The volume to be applied can be of several ml depending on the type of pipette used. MiniColumn efficiency typically replicates performance of larger columns packed according to manufacturer’s recommendation.

MiniColumns are ready-to-use and may be arranged on the base plate in any specific order, according to the requirements of the proposed resin screening or methods development experiment. MediaScout® MiniColumn arrays are delivered with capping mats and equilibrated in 20 % ethanol, with or without 0.15 M sodium chloride, depending on the particular resin. The array may be resealed with the cap mat after use and stored under apprpriate conditions, e.g.at room temperature when equilibrated with 20% ethanol. Column position may be rearranged by the user should the need arise. Individual column rows may also be ordered separately for replacement purposes.

Set pipette into dispense mode. Regulate the speed to about 0.5 ml per minute

These MediaScout MiniColumns are of 5mm internal diameter and up to 10mm bed height.
They each contain up to 0.2 ml of the chromatography resins, packed to defined density, corrected for each individual resin material, in order to replicate conditions in larger columns.

The liquid reservoir volume is:
230µl with 200 µl resin bed
280µl with 100 µl resin bed
315µl with 50 µl resin bed

Columns are run stepwise, and are designed for resin screening and for the early stages of method scouting.
MiniColumn efficiency typically replicates that of larger columns packed according to manufacturer’s recommendation.

RoboColumn


Robot-operated HTS tool in a 96 array format

These MediaScout MiniColumns are of 5mm internal diameter and up to 30mm bedheight.
They each contain up to 0.6 ml of the chromatography resins, packed to defined density, corrected for each individual resin material, in order to replicate conditions in larger columns.
Longer RoboColumns up to 50mm bed height are under development.
New versions of 8mm (24 units on one 96 array plate) and 11.3mm ID (8 units on one 96 array plate) both with 50mm length are also under development.

Columns are run in principle stepwise, but could be run continuously while the flow is controlled by a positive displacement system and applied by a fixed tip needle trough a special needle adaptor. They are designed for comprehensive resin screening and for the early stages of method scouting in an advanced automated way. The volume to be applied can be several ml depending on the type of system used.
MiniColumn efficiency typically replicates that of larger columns packed according to manufacturer’s recommendation.
