Chromatography of particulate containing materials with a NEW Process Column Design
Proxcys, www.proxcys.com have introduced a column system "Axcis", scaleable from lab to manufacturing use with any process resin, which can operate with unclarified feed material without blocking the inlet screen. Tangential circulation of the load material keeps the screen clear whilst pressure allows normal load through the screen onto the packed bed.

The design is compatible with any traditional resin and adsorption mode of operation. Columns are packed to high efficiency into a stable radial configuration. This offers additional advantages in terms of convenient scaleability and reduced footprint in manufacturing facilities.

Solving scale-up and solubility challengesin liquid chromatography
Dynamic Extractions announce a range of High Performance CCC systems that scale from analytical to kilo quantities.

"Counter Current Chromatography" is in principle a very simple technique based on partitioning. This means it is predictable and reproducible.

Separations of complex mixtures are achieved without the need for solid stationary phases which would not tolerate high levels solids or particulates in the crude injection. By using two liquid phases, one stationary the other mobile, products with low solubility can be processed and isolated to high purity.
Due to the absence of a packed column
  • Methods are developed at analytical scale (mgs) and can be directly scaled up to kilogram scale
  • There is no packed bed to clean or validate.
  • This powerful technique will tolerate the presence of cell debris and other particulate impurities, so feedstock clarification is not required!
In principle this technique is possible with any mode which uses immiscible solvents, and is currently best suited to small molecules or peptides, rather than solvent sensitive products such as proteins etc. Any method using RPC or NPC can be adapted to this technique.

Novexin’s proprietary NVoy technology offers substantial improvements in many aspects of bioproduct production and analysis. It can be used to maintain yield and improves recovery for proteins, viruses and other bioactive preparations.

This unique polymer chemistry reduces protein aggregation and non-specific binding, enhances protein stability, maintaining proteins in solution at high concentration and avoiding the use of detergents. The technology also improves refold yield and refolded protein quality.

The polymer is easily removed from proteins (by ion exchange, hydrophobic interaction or affinity chromatography) and is compatible with commonly used analytical techniques such as SDS-PAGE, mass spectrometry, circular dichroism and immunoassay.

Chirobiotic®T for peptide analysis

Closely related peptides that are different from each other because of a particular single amino acid or because of the chirality of an amino acid either within the peptide or at its terminus are of great importance in drug discovery. Current LC analysis usually employs reversed phase (C18) methods which often need ion pairing reagents that modify the column and are not MS compatible, or ion exchange LC that uses buffers that are also generally not MS compatible.
LC/MS compatibility and improved alternative selectivity:

The resolution of small peptide sequences can now be enhanced utilising new macrocyclic glycopeptide chiral stationary phases. Chirobiotic mobile phases are generally acetonitrile-water or acetonitrile-buffer based systems that are LC/MS compatible, enabling higher sensitivity than UV detection. More importantly, Chirobiotic phases provide different selectivity, higher resolution and higher capacity when compared to conventional C18, ion pairing and ion exchange methods. Peptides up to 16 amino acids have been separated and current development is underway for peptides to 30 residues in length.
